  • Overview

The Nascent Transcriptomics Core and Weinstock laboratory at Harvard Medical School are seeking a shared Research Technician to join our team.  Analysis of nascent RNA – RNA that is actively being synthesized by RNA polymerase – can reveal details of the cell's transcriptional responses to drugs and other perturbations in a way that other RNA sequencing methods can miss. Further, nascent RNA-seq is quickly becoming a method of choice for the identification of regulatory elements such as enhancers.  To support the growing interest in the nascent transcriptome across the research community, the core aims to make high-quality nascent RNA library construction and analysis available across the HMS community.  The Weinstock laboratory partners with the Core to develop strategies for applying nascent transcriptomics to the biology and targeting of lymphoid malignancies.

The Research Technician will work with the Weinstock laboratory and the Core and report to both. Responsibilities will include: the construction of nascent RNA libraries, optimization of existing methods, continued innovation of new techniques and application of this technology to biologic questions relevant to lymphomas and leukemias.  This is an opportunity to learn cutting-edge NGS methods, gain experience with laboratory automation (Agilent Bravo) and provide a valuable, collaborative service to scientists across Harvard Medical School and the greater Boston area.

    Primary Duties

    • Isolate and analyze RNA from user samples according to established protocols
    • Construct NGS libraries manually and using the Bravo automation platform
    • Perform quality control (QC) on libraries using qPCR, TapeStation, gel electrophoresis, etc.
    • Keep accurate, up to date records of all samples and experiments
    • Serve as an interface between the core and Weinstock laboratory members
    • Participate in Weinstock laboratory meetings and career development opportunities

    Secondary Duties

    • Assist with upkeep of laboratory records and equipment as needed
    • Perform other laboratory duties as assigned by the core director

This is a unique opportunity to learn state-of-the art automation and NGS library construction strategies, and to contribute to multiple projects of direct biomedical relevance.  This position demands exceptional attention to detail and the ability to precisely follow and interpret complex and lengthy molecular biology protocols.  The successful applicant will be well organized, focused, and able to work efficiently on multiple tasks with deadlines.  The work environment is highly collaborative, thus optimal candidates will work well as part of a team, and possess strong interpersonal and communication skills. Above all, successful candidates will display a commitment to performing high quality research with advanced technology.
